Purification and Microencapsulation of Laccase for Application in Bio fuel cells

Description
Laccase (Lac) as a copper-containing oxidase is widely distributed in fungi, higher plants, and some bacteria, which cancatalyze the oxidation of many phenolic and non-phenolic compounds accompanied by the reduction of oxygen. Lac has been applied to many industrial processes including decolourization of dyes, pulp delignification, oxidation of organic pollutants in wastewater, microbial transformation of natural products, and the development of biosensors for determination of phenols and their derivatives in blood, wine, tea, fruit juice, and oil. This work aims to investigate the production and enhancement of laccase productivity, by selected white rot fungus(pleurotus ostreatus), through solid state fermentation, study optimum ph, optimum temperature, production media , inoculums size, investigate enhancement of laccase activity at low cost via different strategies, such as inducer addition and lignocellulosic wastes ,study the effect of gamma radiation on production and activity of laccase enzyme ,immobilize the enzyme by microencapsulation method ,purification and characterisation of laccases from the selected fungus , study the kinetic parameters of laccase enzyme and try to apply the purified enzyme in the construction of bio fuel cell
