Description
This thesis explores the effects of pollutants from the industry in the English Channel, in particular radionuclides, on the early stages of development of the Japanese oyster Crassostrea gigas. Original experimental devices were designed to study in laboratory controlled conditions the growth of a large number of larvae and spats. Emphasis was given to a joint use of stress markers at integrated and molecular levels. First exposures to non-radioactive pollutants revealed a highly sensitive response of the growth of juveniles exposed to a metal (zinc). Thereafter, juveniles were exposed for several weeks to dose rates of ionizing radiation and radionuclide activities several orders of magnitude higher than the levels likely to be encountered in the marine environment. An eco-geno-toxicological approach contributed to the understanding of the effects of ionizing radiations on DNA. The results strongly suggest that biochemical mechanisms act efficiently against radio-induced damages as early as the first days of life of the Japanese oyster. It still remains a lack of information about the long-term effects of DNA damages to higher level of biological organization. (author)
Abstract (French)
Ce travail de these porte sur les effets chroniques de contaminants industriels en mer de la Manche, dont les radionucleides, sur les stades precoces de developpement de l'huitre japonaise Crassostrea gigas. La conception de dispositifs experimentaux originaux a permis d'etudier en laboratoire la croissance d'un grand nombre de larves et de naissains. L'accent a ete mis sur l'utilisation conjointe des marqueurs de stress a des niveaux integres et moleculaires. Les premieres expositions a des contaminants non radioactifs ont mis en evidence une reponse sensible et precoce de la croissance des juveniles vis-a-vis d'un metal (zinc). Par la suite, les juveniles ont ete exposes plusieurs semaines a des debits de doses de rayonnements ionisants et des activites en radionucleide superieurs de plusieurs ordres de grandeur aux niveaux susceptibles d'etre rencontres dans l'environnement marin. Une approche eco-genotoxicologique a permis d'apporter des elements de reponse quant aux effets des rayonnements ionisants sur l'ADN. Les resultats suggerent fortement que des mecanismes biochimiques agissent efficacement contre les dommages radio-induits des les premiers jours de vie de l'huitre japonaise. Il reste encore des interrogations quant aux effets a long terme des dommages a l'ADN sur les niveaux d'organisation biologique superieurs. (auteur)
