A new method for labeling fibrinogen with 99m-technetium

  • 2. University of Basel (Switzerland)

Description

Labeling of fibrinogen with 99m-Tc still is a problem, because of the pH-sensitivy of the protein. If the solution is too acid denaturation will take place, if it is too alkaline there will be hydrolysis. Therefore a control of the labeling rate is not sufficient, there must also be a control of the clottability of the fibrinogen. The pertechnetate-ion which is obtained by elution from a generator is reduced by tin(II) chloride in a solution of citric acid which is neutralized with sodium hydrogencarbonate. In this work the clottability of fibrinogen was tested by the method of Clauss, the labeling rate was determined by column chromatography
