The 14C-monomethylamino-antipyrine breath test as in vivo parameter for characterizing the induction of the drug catabolizing enzyme system in the guinea pig

The aim of these investigations was to help clarify the following questions: 1) Does MAAP, following 14C labelling of the exocyclic aminomethyl group, offer a suitable substrate for a breath test in guinea pigs. 2) Which procedures for evaluating the 14C exhalation curves of the breath test are especially valid. 3) Can an induction of the drug catabolizing enzyme system following pre-treatment with various inducing substances be detected by the 14C-MAAP breath test. 4) Do inducer-specific differences arise in response to the 14C-MAAP breath test by which the inducers can be characterized. 5) Is monomethylamino-antipyrine similar to amidopyrine in that it is a suitable independent in vivo parameter for the drug metasbolizing enzyme system in the liver of guinea pigs. Die Versuche dieser Arbeit sollten zur Klaerung folgender Fragen beitragen: 1. Eignet sich MAAP nach Markierung an der exozyklischen Aminomethylgruppe mit 14C als Substrat fuer einen Atemtest beim Meerschweinchen. 2. Welche Auswertungsverfahren fuer die 14CO2-Exhalationskurven des Atemtests sind besonders aussagekraeftig. 3. Ist eine Induktion des arzneimittelabbauenden Enzymsystems nach Vorbehandlung mit verschiedenen induzierenden Substanzen durch den 14C-MAAP-Atemtest nachzuweisen. 4. Bestehen Induktor-spezifische Unterschiede im Ansprechen des 14C-MAAP-Atemtests, durch die die Induktoren charakterisiert werden koennen. 5. Eignet sich Monomethylaminoantipyrin aehnlich wie Amidopyrin als eigenstaendiger In-vivo-Parameter des arzneimittelmetabolisierenden Enzymsystems in der Leber des Meerschweinchens. (orig./MG)
