Experimental basis for the combination of irradiation with cytotoxic drugs

Description
For the treatment of therapy resistant tumours the combination of irradiation with chemical substances, especially cytotoxic drugs, can have advantages. In order to obtain supraadditive effects it is reasonable to know the mechanism of interaction. In this respect interactions are very important on the level of intracellular recovery processes, of repopulation, and of hypoxic cells. Substances have been discussed which interact with ionizing radiation on these levels. Especially the modification of recovery processes can increase the radiation effects efficiently. In this connection substances like actinomycin D, bleomycin, adriamycin, and newer drugs like aclacinomycin are of interest. For such a treatment tumours with a high capacity of recovery must be considered. An individual selection would have great advantages therefore. (orig.)
Abstract (German)
Fuer die Behandlung therapieresistenter Tumoren kann die Kombination von Bestrahlung mit chemischen Substanzen, insbesondere Zytostatika, eine Verbesserung ergeben. Um ueberadditive Effekte zu erhalten, ist es sinnvoll, den Mechanismus der Interaktion zu kennen. Dabei sind Interaktionen auf dem Niveau der intrazellulaeren Erholungsvorgaenge, der Repopulierung und hypoxischer Zellen besonders wichtig. Es werden Substanzen besprochen, die auf diesen Niveaus zu Interaktionen mit ionisierenden Strahlen fuehren. Insbesondere die Modifizierung von Erholungsvorgaengen kann zu einer erheblichen Steigerung der Strahleneffekte fuehren. In diesem Zusammenhang sind Substanzen wie das Actinomycin D, Bleomycin, Adriamycin und neuere Stoffe wie das Aclacinomycin von Interesse. Fuer eine derartige Behandlung kommen vor allem solche Tumoren in Betracht, die eine hohe intrazellulaere Erholungskapazitaet haben. Eine individuelle Auswahl waere daher von grossem Vorteil.
