Effect of platelet activating factor (PAF) on rat ovarian granulosa cell secretion of E2

Description
Objective To study the possible mechanism of augmentation of rat ovarian granulosa cell secretion of E2 in vitro by PAF. Methods: Rat ovarian granulosa cells were recruited and cultured under different condifions: 1) no stimulant added 2) FSH 1μg/ml added 3) PAF 1μg/ml added 4) forscolin 100ng/ml added 5) phorbol-12-myristate 13-acetate(PMA) 100ng/ml added. 6) PAF and FSH combined 7)PAF and forscolin combined 8) PAF and PMA combined. The E2 concentrations in various cultures were determined with RIA. Results: Each of the four stimulants PAF, FSH, forscolin, PMA could induce hypersecretion of E2 by rat ovarian granulosa cells (P<0.05). Combined PAF with the other three stimulants could furthar augment the hypersecretion (P<0.01). Conclusion: PAF could stimulate the E2 secretion from cultured rat ovarian granulosa cells via pka and pkc signa transmission pathways. (authors)
