Inhibition of Rad51 via RNA interference leads to MMC sensitisation but not radiation sensitisation in CHO cells

Description
This study was dedicated to examining the effect of inhibition of Rad51 protein through RNA interference on the sensitivity to mitomycin C and ionising radiation in a cell line with intact NHEJ (CHO-K1) as compared with a cell line with defective NHEJ (xrs5). In adult cells inhibition of Rad51 protein through RNA interference has no effect on radiation sensitivity. It follows that Rad51 only plays a minor role in the repair of DNA double strand breaks in cells with intact NHEJ
Abstract (German)
In dieser Arbeit wurde untersucht, welchen Effekt die Hemmung des Rad51-Proteins via RNA-Interferenz auf eine Zelllinie mit intaktem NHEJ (CHO-K1) im Vergleich zu einer Linie mit defektem NHEJ (xrs5) auf die Empfindlichkeit gegenueber Mitomycin C und ionisierende Bestrahlung hat. Die Inhibition des RAD51-Proteins durch RNA-Interferenz hat in adulten Zellen keinen Effekt auf die Strahlenempfindlichkeit. Folglich spielt Rad51 nur eine untergeordnete Rolle in der Reparatur von DNA-Doppelstrangbruechen in Zellen mit funktionellem NHEJ. (orig.)Additional details
