Radiosensitizing effect of MISO-porphyrin-DTTA on Lewis lung cancer

Description
Objective: To investigate the radiation-sensitizing effect of MISO-porphyrin-DTTA on animal model. Methods: CsT BL/6 mice transplanted with Lewis lung cancel were div/dod into 7 groups randomly: control; hypoxia alone; MISO-porphyrin-UITA alone; radiation alone; radiation + MISO-porphyrin-DTTA; hypoxie radiation; and hypoxie radiation + MISO-porhytin-DTTA. For hypoxia alone, the blood flow in the minor beating leg was interrupted for 10 min. MISO-porphyrin-DTTA alone group was further divided into 2 sub-groups according to the dose of MISO-porphyrin-DTFA with 8.58 and 17.15 mg/kg, both in single intraperitoneal injection. Radiation alone group was divided into 3 sub-groups according to different radiation single dose (7.5, 15.0, 25.0 Gy). Radiation + MISO-porphyrin-DTFA group was divided into 6 sub-groups. Hypoxic radiation + MISO-porphyrin-DTFA group was divided into 6 sub-groups, each group including 8 mice. Method of tumor growth delay was used in the expefimem. Results: Mean sensitizing enhancer ratio on oxygen was 1.62 and 1.89, with 1.57 and 1.77 on hypoxia, according to 8.58 or 17.15 mg/kg MISO-Porphyrin-DTTA when the tumor volume was 4 times of its initial volume. Conclusion: MISO-porphyrin-DTIA on dose of 8.58 or 17.15 mg/kg do have radiation-sensitizing effect on Lewis lung cancer mad are avirulent to C57BL/6 mice. (authors)
