Incorporation of uridine-H3 into healthy and tobacco necrosis virus-infected mesophyll cells of Chenopodium amaranticolor

Tritiated uridine was selectively incorporated into the nucleus, nucleolus and cytoplasm of actinomycin D-treated Chenopodium amaranticolor cells locally infected with a strain of tobacco necrosis virus (TNV), 3 days after inoculation. Healthy cells did not show such an incorporation. Chloroplasts, in both types of cells, were free of label. Treatment with pancreatic ribonuclease removed the label completely in the majority of nuclei and nucleoli of infected cells. Since infectivity tests showed that AMD treatment increased virus multiplication by 10-12%, it is conceivable to think that the incorporation observed was due to virus synthesis. Preliminary infectivity experiments also showed that treatment of the cells with cycloheximide inhibited virus multiplication up to 80%, while chloramphenicol increased such multiplication. Our results lead to the conclusion that nucleus, nucleolus and cytoplasm but not chloroplasts are the sites involved in the synthesis of TNV. (orig.)

Uridin-H3 wurde in Zellen von Chenopodium amaranticolor, die mit TNV infiziert und nach drei Tagen mit Actinomycin D behandelt wurden, selektiv in Kern, Nucleolus und Cytoplasma eingebaut. In gesunden Zellen erfolgte kein Einbau. In den Chloroplasten wurde Uridin in beiden Faellen nicht eingebaut. Eine Behandlung mit Pankreas-Ribonuclease fuehrte in der Mehrzahl der Kerne und Nucleoli infizierter Zellen zum Verschwinden der Markierung. Die Behandlung mit Actinomycin D steigerte die Virusvermehrung um 10 bis 12%; es ist deshalb moeglich, dass der beobachtete Einbau durch Virussynthese verursacht wurde. Eine Behandlung der Zellen mit Cycloheximid hemmte die Virusvermehrung bis zu 80%; Chloramphenicol steigerte die Virusvermehrung. Die Versuche lassen schliessen, dass Kern, Nucleolus und Cytoplasma (jedoch nicht die Chloroplasten) an der Synthese des TNV beteiligt sind. (orig.)Additional details
