Early response to ionizing radiation: molecular and cellular effects

Description

Ionizing radiation cause by direct and indirect effects via the free radicals resulting from the radiolysis of water, lesions in cellular structure by alterations of the constitutive molecules of cells (lipids, proteins, nucleic acids). Because of the important role of the DNA molecule, radio-induced DNA lesions are more studied. Many cells are able to respond effectively to the DNA lesions in the minutes, the hours, the days which follow the aggression: early response. This respond can lead, in the case of cells irradiated in mitotic activity, to the stop at critical stages of cell cycle, time to activate the systems of repair of genomic lesions. However, when the badly or not repaired DNA lesions durably persist, this state creates a risk of passage of mutations in the daughter cells. The persistence of these lesions can also engage cells towards processes of cellular death by apoptosis or necrosis. In this lecture will be developed different classes of identified lesions, cellular antioxidant defenses, then the various key molecules involved in the early response after irradiation. (author)

Abstract (French)

Les rayonnements ionisants provoquent par des effets directs et indirects via les radicaux libres issus de la radiolyse de l'eau, des lesions dans les structures cellulaires par alterations des molecules constitutives d'une cellule (lipides, proteines, acides nucleiques). En raison du role important de la molecule d'ADN, les lesions radio-induites de l'ADN sont le plus etudiees. De nombreuses cellules sont capables de repondre efficacement aux lesions de l'ADN provoquees par les rayonnements ionisants dans les minutes, les heures, les jours qui suivent l'agression: reponse precoce. Cette reponse peut aboutir, dans le cas de cellules irradiees en activite mitotique, a l'arret a des stades critiques du cycle cellulaire, le temps d'activer les systemes de reparation des lesions genomiques. Cependant, lorsque les lesions de l'ADN mal ou non reparees persistent durablement, cet etat cree un risque de passage de mutations aux cellules filles. La persistance de ces lesions peut aussi engager irreversiblement les cellules vers des processus de mort cellulaire soit par apoptose ou par necrose. Dans ce cours seront developpes les differents classes de lesions identifiees, les defenses antioxydantes cellulaires puis les differentes molecules cles impliquees dans la reponse precoce apres irradiation.
