Radio-immuno assays and ELISA for human leukocyte IFN-α

Description

An enzyme-linked-immunoabsorbent assay (ELISA) was developed for human leukocyte interferon (IFN-α) using monoclonal anti IFN-α as first, and rabbit polyclonal anti-IFN-α as second antibody. The bound antibody is then quantitated by alkaline phosphatase conjugated to goat anti-rabbit-IgG, and is proportional to the amount of IFN-α. Alternatively, [125I] protein A can be used to perform an immunoradiometric assay. The time required for these assays is about 7 hours. For more rapid detection a competitive radioimmunoassay was developed using rabbit polyclonal anti IFN-α antibodies and pure [125I] IFN-α. (Auth.)
