Induction of micronuclei by X radiation and various chemical agents in red blood cells of Pleurodeles waltl. Uptake, release and excretion of one of them: benzo(a)pyrene
Description
The first part of the study is concerned with the effects of X radiation and of various substances dissolved in the breeding water (carbaryl, N-nitrosocarbaryl, benzo(a)pyrene, formol, cafeine, colchicine...) on the formation of micronuclei in red blood cells of Pleurodele larvae. The curves of the dose-effect relationships and of the kinetics of micronuclei formation are established for X radiation and benzo(a)pyrene. In the second part, a scintigraphic study concerning benzo(a)pyrene uptake, release and excretion by larvae, is presented. This study enables the dose-effect curve and the kinetics of micronuclei formation for this substance, to be interpreted. This study must allow the development of a cytogenetic test for the detection of radiomimetic substances in aqueous medium. Pleurodele is proposed as a new animal for the study of genetic toxicology

Nous presentons tout d'abord une etude quantitative des effets des rayons X et de diverses substances dissoutes dans l'eau d'elevage (carbaryl, N-nitrosocarbaryl, benzo(a)pyrene, formol, cafeine, colchicine...) sur la formation des micronoyaux au niveau des globules rouges des larves de Pleurodele. Les courbes de la relation effet-dose et de la cinetique d'apparition des micronoyaux sont etablies pour les rayons X et le benzo(a)pyrene. Dans une seconde partie, nous etudions par scintigraphie, l'absorption, le relargage et l'excretion du benzo(a)pyrene par les larves. Cette etude permet d'interpreter la courbe effet-dose et la cinetique d'apparition des micronoyaux pour cette substance. Cette methode doit permettre la mise au point d'un test cytogenetique pour la detection des substances radiomimetiques en milieu aqueux. Le Pleurodele est propose en tant que nouvel animal pour les etudes de toxicologie genetiqueFiles
