Practical experiences with radioiodine labelling of antibodies - A review

At the present level of protein labelling technique by radioiodine there are a great number of different methods. Each labelling problem can be solved on principle by suitable selection considering wide preservation of native qualities. The lability for natriummetabisulfite, found in previous model experiments of 131I labelling of antibodies, was the reason to modify the classical chloramine T labelling method by using 2-mercaptoethanol as stop reagent. Experimental details of the labelling technique are reported. Experience in solid-phase radioiodination of antibodies by iodogen are represented. The chloramine T method in combination with 2-mercaptoethanol has been the base for preparation and production of a series of high-quality radiotracers for radioimmunological application for 10 years.
