Evaluation of killing effect on MCF-7 cell by thermochemotherapy using the uptake rate of 18F-FDG

Description
To explore the killing effect on MCF-7 cell by thermochemotherapy using 18F-FDG and other indices. In order to observe changes of MCF-7 cell after treatment by thermochemotherapy, the cell survival rate was detected by MTT. The cell apoptosis was checked by flow cytometry and cell ultra structure was observed by transmission electron microscope. The uptake rate of 18F-FDG and the content of lactate dehydrogenase and tumor markers in cell culture medium were employed to assess therapeutic effect of thermochemotherapy. The results showed that thermochemotherapy significantly inhibited the proliferation of MCF-7 cell and induced cell apoptosis. The results of transmission electron microscope demonstrated that the cells treated with thermochemotherapy has a condensation of nuclear chromatin, apoptotic body formation, cytoplasmic vacuolization and organelle loss compare with normal control cells. The content of lactate dehydrogenase in cell culture medium did not change significantly after 24 h thermochemotherapy, while the content of glucose was increase and resulted in a decrease for the uptake rate of 18F-FDG. Thermochemotherapy has a great influence on glucose metabolism of cells. The uptake rate of 18F-FDG in cell culture medium could be a better index than the content of lactate dehydrogenase and other indices in the evaluation of therapeutic effect of thermochemotherapy.
