A beta-endorphin radioimmunoassay: specificity studies

Description

This note describes the technical details of a cerebrospinal fluid (CSF) B-Endorphin (B-End) radioimmunoassay (RIA). We used an antiserum raised in rabbits against human B-End which cross-reacs 100% with B-Lipotrophin (B-Lph). Thus, filtration chromatography is used to separate both peptides. The assay is sensitive (limit detection=14 pmoles/l), reproducible (the intra and inter assay coefficients of variation are 5 and 6% respectively). Specificity studies are performed to verify the cross-reactions with other opioid peptides and the non specific reactions with the biological fluid (CSF). In order to evaluate the effects of iodine-containing contrast media on the RIA, additional standard curves were analyzed in the presence of varying concentrations of two contrast materials

Abstract (French)

Cette note decrit une methode radioimmunologique (RIA) de dosage de la beta-endorphine (B-End) dans le liquide cephalo-rachidien (LCR) humain. L'antiserum anti B-End utilise dans ce dosage croise a 100% avec la B-lipotrophine (B-Lph), aussi les deux peptides sont separes par chromatographie par permeation de gel. La methode est sensible (limite de sensibilite = 14 pmoles/l), reproductible (coefficients de variation intra et inter-essai de 5 et 6%). L'importance des etudes de specificite sera montree en etudiant les reactions croisees avec d'autres peptides opioides, et les reactions non specifiques avec le milieu biologique teste (LCR). Une etude des interferences in vitro avec des substances iodees sera presentee
