Formulation of macroaggregated albumin (MAA) kit for lung perfusion

Description

Formulation and quality control of Macroaggregated Albumin (MAA) kit to be labelled with technetium-99m (99mTc) have been carried out. These 99mTc-MAA kit to be use for pulmonary perfusion Lyophilized MAA kit was prepared aseptically and analyzed for its pH, radiochemical purity, amount and particle size, sterility, pyrogenity, biodistribution test. The labelled MAA with 99mTc were analyzed by paper chromatography method with 85 % methanol as eluent and Whatman-1 paper as stationary phase, while the sterility test was performed using fluid thioglycolate (FTG) and tryptone soy broth (TSB). In vivo study was carried out in male mice. The experimental results showed that the pH of the kit was 5, radiochemical purity 99.4 %, the sterility test showed no microorganisms growth in culture media, no significant increase in temperature of experimental animals. Biodistribution in male mice showed that the distributed particles in the lung were 84.85 % while the percentage of the particles in the liver was 1.54 %. The ratio of particles in the lungs to the liver is 55:1. The results showed that the MAA kit made was eligible for radiopharmaceutical injection. (author)
