News and views on tumor markers: The use of radioactive antibodies against cell-bound antigens

It was doubtless due to the phenomenal progress in the field of tumor immunology that took place during the last 20 years, that today we dispose not only of highly sensitive immunological tests like the RIA or EIA, but also of most specific reagents like monospecific polyclonal and monoclonal antibodies. In this context the discovery in human carcinomas of tumor-associated antigens was of prime importance, especially since some of them were found to have clinical relevance as so-called tumor markers. It has been shown that there is a direct correlation between the absolute tumor burden and the blood concentration of these substances. Based on animal models a new technology for tumor and metastases detection was developed in recent years, that used polyvalent or monoclonal antibodies prepared against tumor-associated antigens. This technique called radioimmuno-detection (RAID), especially in the hands of experts, may be superior in many instances to conventional radiology, radionuclide scanning or ultra-sonographic techniques. Abstract (German)
Dank des grossen Fortschritts der Tumorimmunologie in den letzten zwanzig Jahren verfuegen wir heute nicht nur ueber hoechst sensible immunologische Nachweismethoden, wie die RIA oder EIA, sondern auch ueber hochspezifische Reagenzien in Form von monospezifischen polyklonalen und monoklonalen Antikoerpern. In diesem Zusammenhang war die Entdeckung von karzinofoetalen Antigenen in menschlichen Tumoren von ganz besonderer Bedeutung, da einige sich fuer die Klinik als sehr nuetzlich erwiesen haben. Es hat sich naemlich gezeigt, dass diese als Tumormarker bezeichneten Substanzen eine direkte Korrelation zwischen der Serumkonzentration und dem absoluten Tumorvolumen aufweisen. In den letzten Jahren ist nun eine neue Technologie entwickelt worden, die radioaktiv markierte polyklonale und/oder monoklonale Antikoerper, die gegen die Markersubstanzen gerichtet sind, zur Tumor- oder Metastasenlokalisation benutzt. Es hat sich gezeigt, dass die Radioimmunodetection (RAID) genannte Methode, in manchen Faellen und von Erfahrenen gebraucht, den klassischen radiologischen, sonographischen und Scanning-Verfahren ueberlegen sein kann.
