Interaction between human platelet factor 4 and protamine sulfate in the presence of a low molecular weight heparin in the rabbit

The effects of different protamine sulfate (PS) doses (15, 30 and 50 mg) upon the kinetics of human platelet factor 4 (PF4) combined with a low molecular weight (LMW) heparin of 7,400 daltons in the rabbit have been studied. The PF4 disappearance after pre-treatment with 1500 APTT units of LMW heparin followed a monoexponential curve with an half-life of 11.03±1.69 minutes. PS at each dosage given 1 minute after LMW heparin and PF4, caused an immediate and deep drop of PF4 circulating level. A second LMW heparin bolus 10 minuts later induced in which 50 mg of PS were given, the PF4 release was only 40% of that obtained in the order two groups of animals
