Preparation, purification and stability of high specific activity 125 sub(I) triiodonthyronine (T3)

Description

A radioiodination of triiodothyronine (T3) by modified chloramine-T method is described. Diiodothyronine (T2) was employed as the starting material, which yields 125I-T3 with specific activities of 110.8 + sub(-) 23.6 TBq/g. Purification by Sephadex G-25 fine column, elution with phosphate buffer in high pH has shown to be convenient, for the separation of the desired products. Studies on the stability of 125I-T3 stored in 50% solution of propylene glycol showed that it is stable for at least 3 moths. The T3 labelled was analysed, using three types of RIA: a) The PEG solution, b) the double antiserum with PEG and c) the solid phase technics. (Author)

Abstract (Portuguese)

Descreve-se a radioiodinacao do triiodotironina (T3) pela modificacao do metodo de cloramina-T. Usou-se o diiodotironina (T2) como material inicial, que produz 125I-T3 com atividades especificas de 110.8+ sub(-) 23.6 TBq/G. Verificou-se que a purificacao por coluna fina de Sephadex G-25, eluicao com tampao de fosfato de alto pH e conveniente para a separacao dos produtos desejados. Os estudos sobre a estabilidade de 125I-T3 estocado em 50% de solucao de propileno glicol mostraram que e estavel por pelo menos 3 meses. O T3 marcado foi analizado usando-se tres tipos de radioimunoensaio: a) solucao PEG, b) duplo soro imunizante com PEG e c) tecnicas de fase solida. (E.G.)
