Effect of free and polymer-bonded phallotoxine on active polymer-bonded antibodies for radio-immunoassay of aldosterone
Description
The preparation of polysaccharide phallotoxine conjugates is described. It is shown that 1. Phallotoxines only bond to F actine and not to G actine, 2. the non-toxic derivatives phallacidine dextrane conjugate, secophalloidine and the monocyclic peptide antamonide have no affinity for actine, 3. besides the toxic derivates phalloidine sulphone and phalboidine sulphoxide B, non-toxic phalloidine sulphoxide A and tribromine phalloidine can also form complexes with F actine. Heat treatment of the phallotoxine actine complex confirms that only toxic derivates stabilize active filaments. Conjugates of phallocidine and phalloidine with sepharose bond purified globular muscle actine which is dissociated by increasing the ionic strength with 0.5M KCl. The affinity chromatography to isolate actine from an impure actine solution with decomposition products is described. (orig.) 891 AJ

Es wird die Darstellung von Polysaccharid-Phallotoxin-Konjugaten beschrieben. Es zeigt sicht, dass: 1. Phallotoxine nur an F-Aktin binden und nicht an G-Aktin, 2. die untoxischen Derivate Phallacidin-Dextran Konjugat, Secophalloidin und das monocyclische Peptid Antamanid zu Aktin keinerlei Affinitaet besitzen, 3. neben den toxischen Derivaten Phalloidinsulfon und Phalloidensulfoxid B auch untoxisches Phalloidinsulfoxid A und Tribromphalloidin mit F-Aktin zur Komplexbildung befaehigt sind. Hitzebehandlung des Phallotoxin-Aktin Komplexes bestaetigt, dass nur toxische Derivate Aktinfilamente stabilisieren. Konjugate des Phallacidin und Phalloidin mit Sepharose binden gereinigtes globulaeres Muskelaktin, das durch Erhoehung der Ionenstaerke mit 0,5M KCl dissoziiert wird. Die Affinitaetschromatographie zur Isolierung von Aktin aus einer mit Zersetzungsprodukten verunreinigten Aktinloesung wird beschrieben. (orig.)Additional details
