Preliminary assessment about genetic diversity, the stability of potential mutants from two varieties of chrysanthemum MORIFOLIUM RAMAT. (Bronze Doa and Purple Farm) via gamma irradiation

Description
Gamma ray from 60Co source was used to irradiate on Chrysanthemun morifolium Ramat (Bronze and Purple chrysanthemum), through that selected a number of potential mutants for choosing new cultivars in future. The experimental result showed that LD50 for the Bronze Doa variety was 50 Gy and Purple Farm variety was 100 Gy, depending on these results we determined the dose for irradiation on Bonze chrysanthemum was 10-20-30-40-50 Gy, Purple chrysanthemum was 20-40-60-80-100 Gy. Irradiated in vitro artificial seeds were transferred into fresh MS medium and placed in the growth room with three replications at each of dose. In this study, on farm, through screening 18 phenotypic mutants of both chrysanthemums were recorded and collected including 6 potential mutants that selected for next research based on their phenotypic differences to the originals, their aesthetic and low mosaic. They were morphologically stable on farm through 3 series of growing on farm at M1V3 generation. The genetic tool was random amplified polymorphic DNA (RAPD), the genetic homologous coefficients of 2 potential mutants from Bronze Chrysanthemum were 0.803 and 0.869, while of 4 ones from Purple Farm were a range from 0.672 to 0.902. They possessed the high stabilities through in vitro generations (98%-100%). All of these criteria show that, these mutant lines were already to be registered as temporary cultivars/varieties.
