Polypeptide mapping of citrus tristeza virus strains
Description
Two coat proteins were present when purified citrus tristeza virus (CTV) virions were dissociated and examined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Purified CTV was labelled with 125I using the chloramine T procedure. The larger coat protein (CP1), about 23 000 MW, was present in greater quantity than the smaller coat protein (CP2), about 21 000 MW. Coat proteins from several aphid-transmitted Florida CTV isolates having different biological activities were compared by polypeptide mapping on SDS-PAGE after dissociation and proteclytic digestion. The polypeptide maps revealed similar patterns for all isolates with only minor differences between some isolates. Polypeptide mapping of CTV coat proteins would not be a useful general technique to detect strain differences
