In vivo induction of cytotoxic T lymphocytes (CTL) in H-2K/sup b/ mutant system and cross-reactivity (CR) analysis at the level of narrow-specific CTL clones (NSC)

Anti wild-type (B6) H-2K/sup b/ mutants (bm) CTL were induced in the regional lymph nodes by 2 injections (with 2 wk interval) of bm mice into foot-pads with B6 irradiated splenocytes (IRS). CTL were tested 7d after the boost including 3d precultivation in monoculture (required for high CTL activity in bm). Active bm4 CTL were induciable in vivo but not in mixed lymphocyte culture (MLC) while bm1 and bm3 were equally active in both models. Specific suppressor T cells (SSTC) were induced in vivo by i.v. injection of 108 B6 IRS. Only bm3 showed suppression of anti-B6 MLC. In vivo induced bm3 CTL were cloned with B6 IRS stimulators and rat interleukine 2. Of 10 Thy1.2+ NSC 2 displayed CR to allogeneic target cells (TC): 1-st lysed K/sup k/TC (B10.A(2R) and the 2-nd-K/sup d/TC (B10D2. (R101)). Implications:a the epitopes detected on H-2K/sup b/ by CTL and SSTC do not coincide, b the in vivo and in vitro induced CTL are not identical, c NSC show various CR to H-2 antigens, possibly because of receptor's affinity difference
