Preparation of Cisplatin for Chemotherapy Application of Cancer

Description
Cis-Plain was prepared in two different methods, and completely characterized by recording IR, UV, Visible spectrum and powder X-ray diffraction (XRD) of the prepared complex. The results were completely conform to those of international publications. Kurnakov test was carried on the water solution of the prepared complex, which distinguish between the two isomers Cis Platin and Trans- Platin, the test gave positive result, this test is carried on by adding Theo-Urea solution (H2NCSNH2) to the complex and heating the final solution, it will give dark yellow colored solution in the case of Cis- Platin complex. Sterile pharmaceutical kits were prepared of the Cis-Platin compound, with concentration of 500mg/ 100 ml of physiologic serum. Cellular studies were carried on (tumor cells of human lungs) to illustrate the level of the effect of Cis-Platin compound on the growth of A549 cells (concentration- time), this study was performed on the compound prepared in the two methods C1, C2. The results were compared with those of commercial Cis-Platin C3. A549 cells treated for 24 hours with increasing concentrations (1,4,10,30,50,100 μg ) of cisplatin compounds ( C1 , C2. C3) showed raised decreasing in their vitality starting from concentraction (10 μg) for three compounds in comparison with blank 100% .On the otherwise , cells treated for 72 hours with (10 μg) concentration of three compounds showed highly decreasing in their vitality which execute to parity complete cells death . and from this results the concentration of (IC50) for the compound was definite with the value 25 μg for C1 compound , 10 μg for C2 compound, and 4 μg for C3 compound which upon it, cisplatincan frustrate growing of half cellular society .cisplatin affect in programmed cellular death was studied for A549 treated cells .Treatment by cisplatin compounds ( C1 , C2. C3) showed direct increasing in cellular death percentage engaged with rising of concentration compound used in treatment in contrast with blank cells group . Annexin V/GFP test which is characteristic for programmed cellular death was used . treatment with cisplatin (7,10 μg) concentrations showed ratio increasing of cells which head for programmed cellular death(author).
