Ku gene mutation of radiosensitive mouse mammary carcinoma-derived cell line SX-9

Objective: To investigate the ku gene status of radiosensitive cell line SX-9 and the effects of ku80 gene mutation on radiosensitivity of SX-9. Methods: After comparing the ku70/80 gene status of SX-9 to SR-1 with molecular cloning, the survival fraction of SX-9 transfected with wild-type human ku80, SX-9 and SR-1 was investigated by clonogenic assay. Results: Radiosensitive mouse mammary carcinoma-derived cell line SX-9 bore mutation in ku80 gene, and its survival fraction after γ-ray irradiation could be restored by transfection of wild-type human ku80 gene. Conclusions: There are two mutation sites in ku80 gene in radiosensitive mouse mammary carcinoma-derived cell line SX-9, which could be the cause of its deficiency in repair of DNA double strand breaks
