Detection of withanolides produced by endophytic fungi isolated from the guitite tree (Acnistus arborescens) using ultra-high-performance liquid chromatography coupled with time-of-flight mass spectrometry

Description

The relationship between the plant and the endophytic fungus goes beyond sharing a space and a means of nutrition. This similarity was confirmed in the secondary metabolites, withanolides, present both in the guitite and in endophytic fungi isolated from it. To detect the presence of the species, the cultivation of inocula from the bark, leaves, and stem was performed in triplicate, followed by extraction and analysis of the extracts using direct infusion mass spectrometry (DIMS) and ultra-high-performance liquid chromatography coupled with time-of-flight mass spectrometry (UPLC-ESI-Q-ToF/MS). Witafisalino I was precisely determined in samples GCH1-2 and GCH1-3, indicating that these are equivalent samples containing the species in the leaves of the shrub. Additionally, in strain GCC7, Fisanolido A was detected, which is a metabolite found in the bark of species from the Solanaceae family. The standard deviation in all measurements was less than 1.0%. This is considered a significant contribution, as guitite is a native species of Costa Rica and has been scarcely analyzed from this perspective. It recognizes new possibilities and contributions to research conducted on the similarity of metabolites with anticancer and antioxidant properties, such as those evaluated in withanolides

Abstract (Spanish)

La relacion entre la planta y el hongo endofito va mas alla de compartir un espacio y un medio de nutricion. Esta similitud se constato en los metabolitos secundarios, witanolidos, presentes tanto en el guitite, como en hongos endofitos aislados del mismo. Para detectar la presencia de las especies, se realizo por triplicado el cultivo de inoculos de la corteza, hojas y tallo, su extraccion y el analisis de los extractos mediante la tecnica de infusion directa acoplada a espectrofotometria de masas (DIMS) y cromatografia liquida de ultra resolucion tambien acoplada a espectrometria de masas de tiempo de vuelo (UPLC-ESI-Q-ToF/MS). Se determinaron con precision mediante sus espectros de masas-masas Witafisalino I en las muestras GCH1-2 y GCH1-3 senalando que son muestras equivalentes que contienen la especie nn las hojas del arbusto. Asimismo, en la cepa GCC7 se detecto Fisanolido A, el cual es un metabolito que se encuentra en la corteza de las especies de la familia Solanaceae. La desviacion estandar en todas las mediciones fue menor al 1,0%. Se considera un aporte significativo, ya que el guitite es una especie nativa de Costa Ria y escasamente analizada desde este enfoque. Reconociendo nuevas posibilidades y aportes a investigaciones realizadas en materia de semejanza de metabolitos con propiedades anticancerigenas y antioxidantes como las evaluadas en los witanolidos.
