Preparation and biodistribution of new neutral myocardial imaging agent 99mTcN(NOEt)2

The preparation methods and biodistribution of 99mTcN(NOEt)2 [bis[N-ethoxy, N-ethyl dithiocarbamato) nitride 99mTc[, a new neutral myocardial imaging agent containing [99mTc≡N[2+ core, are explored. Agents for the preparation of intermediate [99mTc≡N[2+ and ligand NOEt were synthesized. In the presence of reducer and the donor of N, the [99mTc≡N[2+ could be conveniently synthesized at 100 degree C, then NOEt was added, 99mTcN(NOEt)2 formed at room temperature. The partition coefficient of 99mTcN(NOEt)2 was determined and biodistribution of the complex was also studied in rats. Chemical structure of compounds was confirmed by IR, NMR, MS and elemental analysis. The radiochemical purity (RCP) of the complex was evaluated by HPLC and TLC, RCP was over 90%, the complex was stable for 6 hours. Partition coefficient was 427 at pH 7.4. Biodistribution studies demonstrated that this radiopharmaceutical localized selectively in the myocardium of rats, 5 min reached 2.79% ID, while 90 min 1.88% ID. 99mTcN(NOEt)2 exhibits favorable stabilities in vitro and biodistribution properties evident for further studies in human
