X-ray subletal irradiation of murine 3T3 fibroblasts for feeder-layer

Description
Keratinocytes, the main cells of the epidermis, require mitotically inactive but live feeder-layer cells for their survival and proliferation. Four X-ray irradiation doses on 3T3 murine fibroblasts were evaluated for feeder-layer production. Each dose effect was evaluated by cell counting and cellular viability determination (MTT), cell adhesion (plating efficiency) and cell migration (scratch wound healing). Mitotic arrest was suggested by nuclear staining (GIEMSA). An 80 Gy X-ray dose generated enough cell damage to cause mitotic arrest yet keeping metabolically active cells up to 25 days. (author)

Los queratinocitos, las principales celulas de la epidermis, requieren una capa alimentadora de celulas viables mitoticamente inactivadas para su supervivencia y proliferacion in vitro. Cuatro dosis de radiacion con rayos X en fibroblastos murinos 3T3 fueron evaluados para la produccion de capas alimentadoras. Los efectos de las dosis fueron evaluados mediante recuentos celulares y determinacion de la viabilidad (MTT), adhesion (eficiencia de plateo) y migracion celular (scratch wound healing). Ademas, las evidencias encontradas han sugerido el arresto mitotico mediante una tincion nuclear (GIEMSA). Una dosis de radiacion observada correspondiente a 80 Gy genero danos suficientes en la linea celular para causar el arresto mitotico, manteniendo a la vez la actividad metabolica basal durante 25 dias.
