The establishment of an enzyme linked immunosorbent assay for insulin

Description
Two monoclonal antibodies against insulin are applied in two-site enzyme linked immunosorbent assay(ELISA). One of the antibodies is immobilized on the microtiter plate, the other is labelled with HRP. The sensitivity of the assay is 1.7 mIU/L and the recovery is 96.4%-111.1%. The intra- and interassay CV are below 11% and 15.0 % respectively. The correlation coefficient between measured and cliluted values is 0.998 after serial dilutions of the samples using high concentration of insulin. The value for normal samples (n= 38) is 3.6-10.8 mIU/L. Compared with RIA, the correlation coefficient is acceptable (r=0.97). (authors)
