Description
Measurement of the incidence of chromosomal aberrations in the peripheral lymphocytes, though a useful method of biological dosimetry, is techious, difficult to carry out, and time-consuming. It is most appropriate for an assessment of the probability of late damage of low doses up to 20 rad. The many attempts at using biochemical indicators for the absorbed dose have not had satisfactory results so far. Morphological changes especially of the peripheral blood cells and bone narrow cells are a sure sign of a radiolesion. However, there is the problem of quantitative measurement of these processes which has made it impossible so far to elaborate a useful biological dosimetry on this basis. Apart from the analysis of peripheral lymphocytes and leucocythes, there are three new approaches of potential value: 1. Analysis of up to 19 clinical and chemical parameters of the blood serum, - it gives a rough estimate of the probability of lethality of high prognostic value. 2. Observation of the peripheral reticulocyte count - a quick and rather uncomplicated method that does not require specific laboratories. 3. Measurement of the rate of incorporation of labelled iododeoxyuridine in bone narrow cells of radiation-exposed persons or in cell cultures in a medium containing serum of radiation-exposed persons; this method appears promising for the narrow dose range and also raises same questions concerning basic research into intercellular signal substances. Abstract (German)
Von den Moeglichkeiten der biologischen Dosimetrie bietet sich die Messung der Chromosomenaberrationen der peripheren Lymphozyten als brauchbar an. Sie ist allerdings aufwendig, technisch kompliziert und zeitraubend. Sie kann auch bei relativ niedrigen Dosen bis zu etwa 20 rad zur Beurteilung der Wahrscheinlichkeit von Spaetschaeden herangezogen werden. Die zahlreichen Versuche, biochemische Indikatoren fuer die Hoehe einer absorbierten Strahlendosis heranzuziehen, haben sich bisher nicht zu einer brauchbaren biologischen Dosimetrie entwickeln lassen. Die morphologischen Veraenderungen, vor allem der Zellen des peripheren Blutes wie auch des Knochenmarkes sind ein relativ sicheres Indiz fuer einen Strahleninsult. Die Schwierigkeit liegt in der quantitativen Erfassung dieser Veraenderungen, so dass bisher eine nuetzliche, biologische Dosimetrie mit diesen Kriterien nicht aufgebaut werden konnte. Neben der Beurteilung peripherer Lymphozyten und Leukozyten erscheinen drei neuere Ansaetze potentiell brauchbar und vielleicht praeziser. 1) Die Analyse bis zu 19 klinisch-chemischer Parameter des Blutserums ergibt eine grobe Abschaetzung der Wahrscheinlichkeit mit hohem prognostischem Wert eines letalen Ausgangs nach Bestrahlung. 2) Die Beobachtung der Zahl der peripheren Retikulozyten, erscheint als relativ unkompliziertes und nicht an spezielle Laboratorien gebundenes Verfahren, das auch wenig zeitraubend ist. 3) Die Einbaurate von markiertem Joddesoxyuridin in Knochenmarkzellen bestrahlter Individuen bzw. in Kulturzellen, deren Medium Serum von bestrahlten Individuen enthaelt, verspricht vor allem fuer den sehr kleinen Dosisbereich eine nuetzliche Entwicklung und wirft zudem wesentliche Fragen der aktuellen Grundlagenforschung interzellulaerer Signalsubstanzen auf.
