Synthesis and study by nuclear magnetic resonance of deuterated phospholipids
Description
Within the framework of the study of cellular metabolisms and exchanges through biological membranes within which the main compounds are phospholipids, cholesterol, glycolipids and proteins, the author of this research thesis first reports the selective deuteration of the polar head of dipalmitoyl phosphatidylcholine and of the dipalmitoyl phosphatidyl-ethano-amine at the α, β and γ positions. The second part reports the use of NMR for the structural and dynamic study of the phospholipid/cholesterol interaction within model membranes
Abstract (French)
La mise au point d'une methode de preparation rapide et simple de la dipalmitoyl phosphatidylcholine marquee selectivement en α, β et γ du segment choline, nous a permis d'obtenir des quantites importantes (4 g) de ces 3 produits et nous a ainsi donne la possibilite de mener parallelement des etudes en phases lamellaires et en vesicules de l'interaction lipide-cholesterol au niveau de la partie polaire de ce phospholipide par RMN 2H. L'interpretation des resultats obtenus au cours de ce travail a permis, dans un premier temps, de preciser le mecanisme de la relaxation spin-spin T2 du deuterium des phospholipides en vesicules, et dans un deuxieme temps d'etayer un modele possible de l'action du cholesterol sur la partie polaire de ces molecules: l'accroissement de la surface specifique de chaque tete polaire du a la presence de cholesterol permettrait au segment choline d'adopter une orientation plus parallele a la surface de la bicouche, et en augmenterait de ce fait les interactions avec le squelette glycerol. On voit donc au travers de cette etude, que la RMN du deuterium permet une approche a la fois structurale et dynamique des phospholipidesFiles
