Autoradiographical in-vitro examination of the cell proliferation of persisting psoriatic plaques in preferred areas under therapy with ultraviolet light: Photochemotherapy (PUVA) and selective ultraviolet phototherapy (SUP)
Description
The application of ultraviolet rays (PUVA, SUP) brought significant progress in the treatment of psoriasis. The goal of UV-irradiation is to normalise the epidermal growth conditions. The cell cycle of persisting psoriatic plaque in preferred areas was examined under therapy with PUVA and SUP using an autoradiographic in-vitro method, double labelling with 14C- and 3H-thymididin. The results were compared to the known cell-kinetic parameters of psoriasis. The values thus obtained confirm the characteristic changes in the cell cycle of psoriasis. Furthermore, there is a block within the DNA-synthesis phase in the plaques persisting under UV-therapy. The resistence of persisting psoriatic plaques in preferred regions against UV-therapy is regarded as an indication of an extreme disturbance in the proliferation behaviour of the psoriatic skin. Die Anwendung ultravioletter Strahlen (PUVA, SUP) brachte einen wesentlichen Fortschritt in der Behandlung der Psoriasis. Das Ziel der UV-Bestrahlung ist eine Normalisierung der epidermalen Wachstumsverhaeltnisse. An Hand einer autoradiographischen in vitro-Methode, dem Doppelmarkierungsverfahren mit 14C- und 3H-Thymididin, wurde bei 38 Patienten der Zellzyklus der im Bereich der praedilektionsstellen persistierenden psoriatischen Plaques unter der Therapie mit PUVA bzw. SUP untersucht. Die Ergebnisse werden mit den fuer die Psoriasis bekannten zellkinetischen Parametern verglichen. Die ermittelten Werte bestaetigen die charakteristischen Veraenderungen im Zellzyklus der Psoriasis. Darueber hinaus besteht in den unter UV-Therapie persistierenden Plaques ein Block innerhalb der DNA-Synthesephase. Die Resistenz der im Bereich der Praedilektionsstellen persistierenden psoriatischen Plaques gegenueber der UV-Therapie wird als Hinweis fuer eine aeusserst tiefgreifende Stoerung im Proliferationsverhalten der psoriatischen Haut gewertet.
