Methods for hybridization and quantitation of mRNA in individual brain cells

  • 1. Univ. of Arkansas for Medical Sciences, 4301 W. Markham, Little Rock, AR (USA)

Description

In situ hybridization techniques developed to quantitate the levels of specific messenger RNAs (mRNAs) relative to the levels of total polyadenylated (poly-A) mRNA are detailed here together with our protocols for combining immunohistochemical localization of proteins encoded by mRNAs that are localized by in situ hybridization. Specifically, the authors will demonstrate the use of the protocols they have used for in situ hybridization of cDNA, synthetic oligodeoxynucleotides, and RNA probes radiolabelled with either [/sup 3/H], [/sup 35/S], or [/sup 32/P], to paraffin-embedded mammalian tissue preserved with a variety of fixatives
