Description

Preparation of standard for radioimmunoassay. The standard is an essential component in radioimmunoassay. Standard should be of high quality, which in turn is determined by the precise handling and the conditions during preparation. Processing conditions include conditions during separation of serum from red blood cells, preparation of T3 free serum, storage and preparation of standard and drying conditions. Among criteria to determine the quality as standards are calibration results against a reference standard, percentage bound at zero concentrations (Bo/T), non-specific binding (NSB) and parallelism. The experimental values obtained were around 50% for Bo/T, 1% for NSB and the standard concentrations are not significantly different from those of Netria and Amersham. The parallelism test showed that the T3 standard matrix is similar to that of the sample. (author). 3 figs.; 6 refs
