Inhibition of thymidine phosphorylase in vivo provides a rapid method for switching DNA labelling
Description
Uridine blocks the in vivo conversion of thymine to thymidine in Escherichia coli, thus, one can change DNA labels by labelling first with a thymine label (e.g. 14C) and then, at the time of the change, adding 50 μg uridine per ml and thymidine (e.g. 3H). The cells immediately start using the thymidine, ignore the thymine for several generations, and are not affected by the uridine. (orig.)

Uridin blockiert die in-vivo-Umwandlung von Thymin zu Thymidin im Kolibazillus; somit koennen DNA-Markierungen veraendert werden, indem man zuerst mit einer Thyminmarkierung (z.B. 14C) labelt und dann, zur Zeit der Veraenderung, 50 mg Uridin pro ml und Thymidin (z.B. 3H) hinzufuegt. Die Zellen beginnen sofort, das Thymidin zu verwerten, ignorieren das Thymin ueber mehrere Generationen und werden vom Uridin nicht angegriffen. (orig.)Additional details
