Transport and degradation of 2-14C abscicine acid in the coleus rehneltianus berger sprout
Description
1 μg ABA-2-14C aqueous solution was injected into the youngest or into a fully grown leaf of young and older coleus plants. The activity quantity in the various sprout parts is investigated after 2, 24 and 72 h; as well as which labelled substances other than abscisic acid (ABA) occur. The activity in the ethanol extracts was detected with the help of liquid scintillation measurements. Thin layer chromatography and gas chromatography were used to characterize the radioactive substances. The results show that ABA is degraded into three metabolites which are characterized by their relative Rsub(F) values using chromatography with LM 2 as substance 0.2, 0.5a, 0.5b and 0.8. Comparing with the literature shows that it could be 6'-hydroxy methyl ABA, ABA glucoside, phaseic acid and dihydrophaseic acid. Young and old leaves in older plants have the same ability to degrade ABA taking the occurence of the 0.8 metabolite as standard. The degradation in younger plants is firstly very slight. However, within 72 h the ability of ABA degradation is induced in older leaves of younger plants, so that finally the degradation rate is as big as in older plants. Activity is essentially exported by adult leaves of older plants. The basipetal transport is considerably greater than the acropetal one. Results show that mainly ABA is transported in the acropetal test, whereas ABA and fraction 0.5 are transported in the basipetal direction. A reversed transport direction is observed after applying ABA to a young leaf. The transport basipetal is polarized in the first two hours, after 72 hours of transport there is a definite acropetal polarity. (orig./MG)

Juengeren und aelteren Coleus-Pflanzen wird entweder in das juengste oder in ein adultes Blatt 1 μg ABA-2-14C in waessriger Loesung injiziert. Nach 2, 24 und 72 h wird untersucht, wie gross die Aktivitaetsmenge in den verschiedenen Sprossteilen ist und welche markierten Substanzen ausser Abscisinsaeure (ABA) auftreten. Der Nachweis der Aktivitaet in den aethanolischen Extrakten erfolgt mit Hilfe der Fluessigszintillationsmessung. Zur Charakterisierung der radioaktiven Substanzen wird die Duennschichtchromatografie (DC) und die Gaschromatografie (GC) herangezogen. Die Ergebnisse zeigen, dass ABA zu drei Metaboliten abgebaut wird, die nach ihren relativen Rsub(F)-Werten bei Chromatografie mit LM 2 als Substanz 0.2, 0.5a, 0.5b und 0.8 bezeichnet werden. Der Vergleich mit der Literatur zeigt, dass es sich dabei um 6'-Hydroxymethyl-ABA, das ABA-Glucosid, Phaseinsaeure und Dihydrophaseinsaeure handeln koennte. Die Faehigkeit ABA abzubauen ist in jungen und adulten Blaettern aelterer Pflanzen gleich gross, wenn man die Entstehung des Metaboliten 0.8 als Mass dafuer nimmt. In juengeren Pflanzen ist die Abbaurate zunaechst sehr gering. Innerhalb von 72 h wird jedoch in adulten Blaettern juengerer Pflanzen die Faehigkeit ABA abzubauen induziert, so dass schliesslich die Abbaurate ebenso gross ist wie in aelteren Pflanzen. Ein wesentlicher Export von Aktivitaet erfolgt nur aus den adulten Blaettern aelterer Pflanzen. Dabei ueberwiegt der basipetale Transport den akropetalen erheblich. Die Ergebnisse zeigen, dass in akropetaler Richtung offenbar vorwiegend ABA transportiert wird, waehrend in basipetaler Richtung ABA und Fraktion 0.5 transportiert werden. Nach Applikation von ABA ueber ein junges Blatt ist eine Umkehrung in der Transportrichtung zu beobachten. In den ersten zwei Stunden ist der Transport basipetal polarisiert, nach 72 h Transportzeit zeigt sich dagegen eine deutliche akropetale Polaritaet. (orig./MG)
