Investigations on the insulin receptor of isolated fat cells
Description
Fat cells, isolated from the epididymal adipose tissue of rats, were incubed with iodine 125 insulin after previous incubation with various antagonists. By varying the antagonist concentration, it was possible to determine the effect these substances have on the insulin receptor, i.e. the insulin similarity. By varying the preincubation time, toxicity of the test substances could be detected, which pretended repression effects; and by finally verying the incubation time the effects on the receptor via the membrane could be distinguished from direct receptor bindings of the antagonist. Isolierte Fettzellen aus dem Nebenhodenfettgewebe von Ratten wurden nach Praeinkubation mit diversen Antagonisten mit 125J-Insulin inkubiert. Durch Variation der 'Antagonistenkonzentration' konnte die Einwirkung dieser Substanzen auf den Insulinrezeptor, also die 'Insulinaehnlichkeit' bestimmt werden. Durch Variation der Praeinkubationszeit war es moeglich, Verdraengungseffekte vortaeuschende Toxizitaet der Testsubstanzen nachzuweisen, und durch Variation der Inkubationszeit konnten Einwirkungen auf den Rezeptor ueber die Membran von direkten Rezeptorbindungen der 'Antagonisten' unterschieden werden.
