Optimisation of Kanamysin-Tc99m For Infection Imaging

Infectious diseases were responsible for the death of almost 9 million people worldwide in every year. Those infection deaths occur because of late identifications of main infections which occur in very deep parts of the body (deep-seated infection). Kanamycin is an antibiotic belonging to the aminoglycosides, its nature as a broad-spectrum antibiotic allows it to bind to Gram-negative and Gram-positive bacteria by binding to the bacterial ribosome. This study aimed to radiolabel kanamysin with Technicium-99m. Direct labeling were done by adding 5 mg of Kanamysin and 30 μg/ml of SnCl2 with Tc99m solutions. This reaction occurred at pH7 with 30 minutes incubation period at room temperature. Radiochemical purity and stability of the Kanamysin-Tc99m was determined by ascending paper chromatography with 1M NaOH and Acetone as mobile phase. Biodistribution study was done by injecting Kanamysin-Tc99m solution into AZK hairless rats and observed for two hours using Gamma camera. From this study, Kanamysin was successfully radiolabelled with Tc99m at >95 %. Besides that, biodistribution study also showed Kanamysin-Tc99m was excreted normally from the model. These results show that Kanamysin-Tc99m radiopharmaceutical have a potential application for infection diagnosis. Further study using inflammatory model are suggested. (author)
