General approach to the construction of synthetic DNAs

Description

An approach to the construction of extended synthetic DNAs has been developed on the basis of chemical-enzymatic methods of synthesizing polynucleotides. The general strategy of the approach includes the use of temporary recognition sites for macrocleaving restriction endonucleases forming large sections on cleavage in order to facilitate the subcloning and assembly of individual fragments of a gene. The efficacy of the proposed methodology has been demonstrated for the case of the synthesis of a functionally important fragment of the bacteriorhodopsin gene
