Stability 'in vivo' and 'in vitro' of the diisopropyl-IDA sup(99m)Tc

The 'in vitro' and 'in vivo' stability of the diisopropyl (IDA-3) kit in solution and of the molecule labelled with sup(99m)Tc was studied. The following items were studied a) shelf life of the kit in solution b) 'in vitro' stability of the labelled molecule c) distribution and elimination kinetics d) identification and behaviour of the excretion products. Chromatographic control shows 95% yield of labelled product after 20 days storage at 4 deg C and 92% yield after 30 days in the same conditions; biological distribution was good in both cases. Regarding the 'in vitro' stability, no alteration was observed within the 4 hours after its preparation. The stability of the complex was determined by the reaction between sup(99m)TcIDA-3 and DTPA. Analysis by electrophoresis shows the complex to be stable even with DTPA concentrations 5 times higher. Kinetics of distribution and elimination of the labelled molecule, in animals, were afterwards confirmed by sequential scintigraphic images in humans. The excretion of unaltered sup(99m)Tc-IDA by biliary and urinary ways was confirmed by cromatographic and electrophoretic analysis of bladder and gall bladder content and by determination of its biological distribution. Se estudio la estabilidad 'in vitro' e 'in vivo' del juego de reactivos de diisopropil-IDA (IDA-3) en solucion y de la molecula marcada con Tc-99m. Se considero: vida util del juego de reactivo en solucion; estabilidad 'in vitro' de la molecula marcada; cinetica de distribucion y eliminacion; identificacion y comportamiento de productos de excrecion. Los resultados por control cromatografico muestran un 95% de rendimiento de marcado a los 20 dias de almacenamiento a 4 grados C y 92% a los 30 dias, habiendose obtenido en ambos casos una buena distribucion biologica. Respecto a la estabilidad de marcado 'in vitro' no se observo alteracion en las 4 horas posteriores a su preparacion. La estimacion de la estabilidad del complejo fue determinada en base a la reaccion de intercambio entre el sup(99m)Tc-IDA-3 y el DTPA. Los resultados electroforeticos muestran que el complejo es estable aun frente a concentraciones 5 veces mayores de DTPA. La cinetica de distribucion y eliminacion de la molecula marcada, en animales, fue confirmada por imagenes centellograficas secuenciales obtenidas en humanos. Se comprobo la excrecion del sup(99m)Tc-IDA-3 sin alteracion a nivel biliar y urinario, hecho constatado por analisis cromatografico, electroforetico y de distribucion biologica del contenido vesicular y de vejiga.
