Effects of 60Co γ-rays irradiation on AT cell cycle progression

Objective: To explore the relationship between cell cycle progression and high radiosensitivity at AT5BIVA cells. Methods: Using flow cytometry to determine cell cycle distribution of AT5BIVA and GM0639 cells from 0-24 h after exposure to 2 Gy 60Co γ-rays and at the 24th hour after exposure to 0-6 Gy 60Co γ-rays. Results: After GM0639 cells were exposed to 2 Gy 60Co γ-rays, the percentage of G2/M cell increased with prolonged time. The percentage of G2/M cell augmented with irradiated dose at the 24th h in GM0639 cells exposed to 0-6 Gy 60Co γ-rays. The percentage of G2/M cells decreased with prolonged time and irradiated dose from 0-24 hour after AT5BIVA cells were exposed to 2 Gy 60Co γ-rays and at the 24th hour after AT5BIVA cells were exposed to 0-6 Gy 60Co γ-rays. Conclusion: The loss of cell cycle check points and the disfunction of DNA repair caused by ATM gene mutation may be responsible for high radiosensitivity of AT5BIVA cells. (author)
