Early kinetics of iodide and pertechnetate: establishment of a compartmental model from data obtained with a scintillation camera

Description
The early thyroid metabolism of iodide may be described by three parameters: the unidirectional clearance C (active iodide transport from the plasma to the thyroid cells), the leakage coefficient ksub(TB) (back-diffusion of thyroid iodide to the plasma) and the organification constant ksub(b) (bonding of iodide to the thyroid proteins). These three parameters were determined, together with their variation under the influence of competing anions such as perchlorate or antithyroid agents such as methyl mercapto imidazol (MMI). After intraveinous injection of the tracer dose (I-131 or Tc-99m) the heart and thyroid activities were measured continuously by external counting with a scintillation camera. The simplest model consistent with facts is a system with 7 compartments for iodide and 6 for pertechnetate. It allows the plasma concentration variation to be determined from the data obtained by external counting, parameters C, ksub(TB) and ksub(b), and MMi-induced organification inhibition
Abstract (French)
Le metabolisme thyroidien precoce de l'iodure peut etre decrit par trois parametres: la clearance unidirectionnelle C (transport actif de l'iodure du plasma vers les cellules thyroidiennes), le coefficient de fuite ksub(TB) (retrodiffusion d'iodure thyroidien vers le plasma), la constante d'organification ksub(b) (liaison de l'iodure aux proteines thyroidiennes). On a determine ces trois parametres, ainsi que leur variation sous l'influence d'anions competiteurs tels que le perchlorate, ou d'antithyroidiens tels que le Methyl Mercapto Imidazol (MMI). Apres injection intraveineuse de la dose traceuse (I-131 ou Tc-99m), on a realise la mesure continue des activites cardiaques et thyroidiennes par comptage externe a l'aide d'une camera a scintillation. Le modele le plus simple compatible avec les donnees est un systeme a 7 compartiments pour l'iodure et 6 pour le pertechnetate. Ce modele permet de determiner: l'evolution de la concentration plasmatique a partir des donnees obtenues par comptage externe, les parametres C, ksub(TB) et ksub(b), l'inhibition de l'organification causee par le MMIAdditional details
