Autoradiographic localization of the synthetic sites of tomato spoted wilt virus and potato virus Y

The biosynthesis sites were investigated of two morfologically different viruses - the Tomato Spotted Wilt Virus (TSWV-spherical particle) and the Potato Virus Y (PVY - long and flexuous particle) in order to discuss the hypothesis of De Zoeten and Schlegel about the relationship between virus morphology and the location of the viral biosynthesis. Samples from uninfected or infected leaves were immersed in distilled water or an aqueous solution and transfered to uridine tritiated solution. After washing in distilled water the samples were fixed, dehydrated and embedded in Epon 812 for electron microscopy conventional techniques. Ultrathin sections were covered with Ilford L-4 photographic emulsion and exposed for two months before photographic development, staining and examinated in the electron microscope. The number of silver grains per unit areas (grain density) in the electronphotomicrographs was used to compare the grains densities of some cells regions of tissues treated or not with AMD. The result indicated the endoplasmic reticulum as the most likely location of the TSWV-RNA replication. The same comparison made with tobacco cells infected with PVY showed that the cytoplasmic area is the most probable site of the PVY-RNA replication. The results obtained seem to show that the rule proposed by De Zoeten and Schlegel cannot be used for all plant viruses because the TSWV replicates in the cytoplasm of infected cell. These viruses seem to be exceptions to that rule. (Author)

Investigou-se os locais de biossintese de dois virus morfologicamente diferentes, ou seja, Virus Vira Cabeca do Tomateiro (VVCT) (particula esferoidal) e Virus Y da batatinha, (particula alongada) visando discutir a relacao forma dos virus e sitio de biossintese. Tecidos foliares de plantas sadias e afetadas com cada um desses virus foram imersos em agua ou solucao aquosa de AMD durante 16 horas, e em seguida transferidas para uma solucao de uridina tritiada e mantidos durante 5 horas. A seguir os especimes foram fixados, desidratados e incluidos em Epon 812 conforme tecnicas usuais para microscopia eletronica. Os cortes ultrafinos foram cobertos com emulsao Ilford L4 e apos 2 meses de exposicao fotografica seguiram-se a revelacao, coloracao, exames e eletrofotomicrografias dos cortes ultrafinos. As contagens de graos de prata reduzida por unidade de area(densidade) em varias organelas indicaram que, sob acao da AMD, o reticulo endoplasmatico da celula infectada com VVCT apresenta relativa alta densidade de graos de prata, indicando que essa extensao do nucleo pode ser o local de biossintese do VVCT. A densidade encontrada na regiao do nucleo, nucleolo das celulas infectadas com PVY e tratadas ou nao com AMD indicam, pela alta reducao de graos de prata, que o RNA viral se replica no citoplasma. Assim, os locais de replicacao do RNA dos dois virus, VVCT e PVY, parecem contrariar a regra estabelecida por De Zoeten e Schlegel que estabelece que o RNA dos virus longos se replica no nucleo e os de forma esferica no citoplasma.
