Hemodynamic and metabolic characterization of orthotopic rat prostate carcinomas using dynamic MRI and proton magnetic resonance spectroscopy

Description
The aim of this study was the noninvasive characterization of prostate carcinoma orthotopically implanted in rats using Gd-DTPA-assisted dynamic MRI (dMRI) and proton magnetic resonance spectroscopy (1H-MRS). After surgical exposure of the prostate, Dunning R3327 orthotopic prostate carcinoma was induced by injecting cells of the MAT-LyLu subline. Six rats were examined 5 and 14 days after tumor induction with dMRI and 1H-MRS at 1.5 T. Six tumor-free rats served as controls. Using an open two-compartment model, the parameters A (amplitude) and kep (exchange rate constants) were calculated from the signal time curves of the dMRI. The relative signal intensities (Cho/Cr) of the resonances of choline (Cho) and the creatine-phosphocreatine complex (Cr) were computed from the MR spectra. Already after 5 days, the tumors in the prostate could be clearly identified based on the decrease in signal intensity to T2w and increase of A and kep. High Cho/Cr levels and resonances of two lipid fractions (Lip1 at 0.8-1.5 ppm and Lip2 at 2.0-2.2 ppm) were observed by MRS in the highly necrotic tumors. The orthotopic rat prostate carcinoma model resembles human prostate carcinoma in regard to MR morphology, dMRI, and 1H-MRS. The noninvasive characterization of perfusion and metabolism makes a comparative examination of different treatment modalities possible. (orig.)

Ziel der Studie war die nichtinvasive Tumorcharakterisierung orthotop implantierter Prostatakarzinome bei Ratten mittels Gd-DTPA-unterstuetzter dynamischer MRT (dMRT) und Protonen-MR-Spektroskopie (1H-MRS). Nach operativer Freilegung der Prostata wurde durch intraprostatische Injektion von Zellen der MAT-Lylu-Sublinie das Dunning-R3327-orthotope Prostatakarzinom induziert. Sechs Ratten wurden 5 und 14 Tage nach Tumorinduktion mit dMRT und 1H-MRS bei 1,5 T untersucht. Sechs tumorfreie Ratten dienten als Referenz. Unter Verwendung eines offenen Zweikompartimentmodells wurden aus den Signal-Zeit-Kurven der dMRT die Parameter A (Amplitude) und kep (Austauschratenkonstante) errechnet. Aus den MR-Spektren wurden die relativen Signalintensitaeten der Resonanzen von Cholin (Cho) und des Kreatin-Phosphokreatin-Komplexes (Cr) berechnet (Cho/Cr). Bereits nach 5 Tagen konnten die Tumoren in der Prostata anhand einer Erniedrigung der Signalintensitaet auf T2w und einer Erhoehung von A und kep deutlich erkannt werden. MR-spektroskopisch wurden bei den stark nekrotischen Tumoren hohe Cho/Cr-Werte sowie Resonanzen zweier Lipidfraktionen (Lip1 bei 0,8-1,5 ppm und Lip2 bei 2,0-2,2 ppm) beobachtet. Das orthotope Prostatakarzinommodell der Ratte aehnelt hinsichtlich MR-Morphologie, dMRT und 1H-MRS dem menschlichen Prostatakarzinom. Die nichtinvasive Charakterisierung von Perfusion und Metabolismus ermoeglicht eine vergleichende Untersuchung verschiedener Therapiemodalitaeten. (orig.)Additional details
