Effects of irradiation on actin in wound neutrophils of rats

Objective: To explore the mechanism of decreased phagocytic and chemotactic function of neutrophils (Neu) and the mechanism of W11-a12's pharmacological action by studying the changes of actin of Neu in wound after total body irradiation of rats. Methods: Cells were collected from incision wounds on back of rats after 6 Gy whole body-irradiation. F-actin in Neu was stained with Fluorescein-phalloidin. Results: There were increased (3-12 h) and decreased (24 h, 48 h) basal levels of F-actin, a lower responsiveness to stimulation and a reduced rate of actin depolymerization after combined radiation injury compared with simple wound group, which were important causes of Neu dysfunction. Conclusion: W11-a12 could improve the functional state of Neu actin, which may contribute to the improved chemotactic and phagocytic function of Neu after administration of this drug
