Irradiation extraction of alkaloids from Arachis hypogaea L. stem and leaf, and the qualitative and quantitative analysis

Description

The aim is to obtain process conditions and parameters of total alkaloids from Arachis hypogaea L. stem and leaf (AHSL) by the irradiation extraction and macroporous resin purification. Total alkaloids of AHSL were extracted by Co-60 γ-ray irradiation at different doses. Total alkaloid content or absorbance value was measured by ultraviolet spectrophotometry. Effects of macroporous resin type and model, and the eluent type and concentration were investigated by static and dynamic adsorption examine. Qualitative and quantitative analysis of AHSL alkaloid were carried out by high performance liquid chromatography. The results showed that a trend of rise first and then fall was observed in total alkaloid content, with the increase of irradiation dose. The highest alkaloid yield was (7.970±0.117)% at irradiation dose of 5 kGy in Co-60 γ-ray irradiation method, which is better than that (1.485±0.061)% in ultrasonic method (t test, p < 0.001). The alkaloids purity can reach 99.99% using AB-8 macroporous resin and elution solvent at 60% methanol elution. In HPLC results, it was found that AHSL alkaloids mainly is choline, and the yield is about 2.61 g·kg-1 AHSL. (authors)
