Nuclear hematology
Description
The determination of erythrocyte life span and the investigation of hematopoietic tissue are discussed. In the former case, the labelling of hemoglobin by glycine 14C and red cells by DFP-32P or Chromium 51 are treated. In the latter case, a tritium injection is used. Refering to the determination of erythrocyte life span, orientation is given about the 'in vitro' method for the labelling of mixed cells of different ages by Chromium-51. The relation among life span changes of red cells and several pathological states is discussed. In the determination of blood platelet life span, orientation is given about the labelling 'in vitro' with Chromium 51. A short review of iron metabolism is presented and methods of investigation used are analysed, such as: evaluation of Iron-59 absorption and excretion; circulation and accumulation of Iron-59 in tissues; Iron-59 uptake by red cells, as well as its distribution through the organs. Three methods are discussed in spleen scintigraphy: use of erythrocytes labelled with Chromium-51 and thermally altered, labelling of red cells by BMHP and use of coloidal Indium-113m. Clinical applications of each method are also described

No estudo da cinetica das populacoes celulares, analisam-se dois metodos: a determinacao da sobrevida das hemacias e a exploracao do tecido hematopoetico. No primeiro, analisam-se a marcacao da hemoglobina pela glicina14C e a dos globulos vermelhos pelo DFP-32P ou pelo Cromo-51. No segundo, emprega-se injecao de tritio. Na determinacao da sobrevida dos eritrocitos, orienta-se sobre o metodo da marcacao 'in vitro' de celulas com idades mistas, detalhando-se a tecnica do emprego do Cromo 51. Correlacionam-se as variacoes da sobrevida das hemacias aos diversos estados patologicos. Na determinacao de sobrevida das plaquetas, orienta-se sobre o metodo da marcacao 'in vitro' com Cromo 51. Faz-se breve revisao do metabolismo do ferro, analisando-se seus metodos de estudo, quais sejam: avaliacao da absorcao e excrecao do Ferro 59, circulacao e acumulo do Ferro 59 nos tecidos; captacao ou aproveitamento do Ferro 59 pelos globulos vermelhos e sua distribuicao pelos orgaos. Na cintigrafia explenica discutem-se tres metodos: emprego de eritrocitos marcados com Cromo 51 e alterados termicamente, marcacao de hemacias pelo BMHP e uso do Indio-113m coloidal.
