Zusammenfassung In einem 3jährigen Langzeitfütterungsversuch mit Ratten wurde an 6 Generationen (456 Tiere) eine Diät, die zu 35% aus bestrahltem (4,5 Mrad) Milchpulver bestand und einen hohen Gehalt an langlebigen freien Radikalen hatte, auf chronische Toxizität geprüft. Folgende Untersuchungen wurden durchgeführt: Wachstum, Futterverwertung, Aufnahme von verdaulichem Eiweiß, hämatologische Untersuchungen, elektrophoretische Trennung und Bestimmung der Serumproteinfraktionen, Gesamtprotein, Serumenzymaktivitäten (GPT, GOT, AP, SDH), Knochenmark‐ und Urinuntersuchungen, Organ‐ (Herz, Leber, Nieren, Milz) und Körpergewichte sowie Sektionsbefunde, histopathologische Bewertung von Organschnitten (Lunge, Leber, Milz, Nieren, Nebennieren) und allen Tumoren, Messungen der Chronaxie sowie Fruchtbarkeit, Reproduktion und Aufzuchtleistung. Signifikante Unterschiede zwischen den Diätgruppen traten lediglich in den Körpergewichten nach 1jähriger Fütterung auf, wahrscheinlich verursacht durch suboptimale Vitaminversorgung der Tiere der Versuchsgruppe. Im 2. Versuchsjahr verschwanden diese Unterschiede wieder. Der Versuch lieferte insgesamt keine Anzeichen einer chronisch toxischen oder cancerogenen Wirkung einer bestrahlten Nahrung mit hohem Gehalt an freien Radikalen. Summary On the wholesomeness of high concentrations of free radicals in irradiated foods A diet containing 35% irradiated milk‐powder (4,5 Mrad) with a high content of long‐lived free radicals was tested with rats using 6 generations (456 animals) in a 3 year longterm feeding study for chronic toxicity. The following parameters were studied: growth, food efficiency, intake of digestible protein, hematological investigations, electrophoretic separation and determination of serum protein fractions, total protein, serum enzyme activities (GPT, GOT, AP, SDH), bone marrow smears, urine tests, organ weights (heart, liver, kidney, spleen), body weights and gross pathology, histopathology of organ slices (lung, liver, spleen, kidney, adrenals) and of all tumors, measurements of chronaxy, fertility, litter size, litter weight and weaning ratio. Significant differences between the diet‐groups were found only with respect to body weights after one year of feeding ‐ probably caused by suboptimal vitamin supply in the experimental group. These differences disappeared during the second year of feeding. This study gave no indication of chronic toxic or carcinogenic effects of an irradiated diet with a high content of free radicals. Résumé Sur l'innocuité de hautes concentrations de radicaux libres dans les denrées alimentaires irradiées An cours d'un assai d'alimentation de longue durée effectué sur des rats pendant 3 ans et sur 6 générations (456 animaux) on a examiné quant à sa toxicité chronique un régime composé de 35% de lait en poudre irradié (4,5 Mrad) et qui présentait une teneur élevée en radicaux libres de longue vie. On a examiné les points suivants: croissance, utilisation de la provende, absorption de protéine digestible, examens hématologiques, séparation par électrophorèse et détermination des fractions protéiniques du sérum, protéine totale, activités des enzymes sériques (GPT, GOT, AP, SDH), examens de la moelle osseuse et de l'urine, poids des organes (coeur, foie, reins, rate) et du corps ainsi que résultats d'autopsie, évaluation histopathologique de sections d'organs (poumons, foie, rate, reins, surrénales) et de toutes les tumeurs, mesures de la chronaxie et de la fertilité, reproduction et performance d'élevage. Après avoir administré le régime irradié pendant une année, des différences significatives ne furent observées que pour le poids du corps; elles sont probablement dues à un approvisionnement suboptimal en vitamines des animaux du groupe testé. Ces différences ont disparue dans la deuxième année. Dans l'ensemble l'essai ne donnait aucune indication d'une action chroniquement toxique on cancérogène d'une nourriture irradiée et d'une haute teneur en radicaux libres. Resumen Sobre la cuestión de la inocuidad saludable de concentraciones elevadas de radicales libres en víveres irradiados En un ensayo de alimentación a plazo largo, que duró 3 años, se contrastó con ratas la toxicidad crónica posible en 6 generaciones (456 animales) de una dieta consistente en un 35% de leche irradiada (4,5 Mrad) y que tenía un contenido elevado en radicales libres de vida larga. Se realizaron los exámenes siguientes: crecimiento, conversión de los alimentos, asimilación de proteína digestible, análisis hematológicos, separación electroforética y valuación de las fracciones seroproteicas, proteína total, actividades seroenzimáticas (GPT, GOT, AP, SDH), análisis de medula ósea y de orina, pesos de vísceras (corazón, hígado, riñones, bazo) y del organismo, así como hallazgos necrópticos, valoración wistopatológica de las cortes de órganos (pulmón, hígado, bazo riñones, adrenes) y todos los tumores, mediciones de la cronaxia, y fertilidad, reproducción y rendimiento de recría. Solo surgieron diferencias significantes entre los grupos de dietas en los pesos en vivo tras alimentación durante 1 año, ocasionadas tal vez por la provisión vitamínica subóptima de los animales del grupo de ensayo. En el 2° año de la experimentación volvieron a desaparecer estas discrepancias. El ensayo no facilitó, en conjunto, ningún detalle de una acción tóxica crónica o cancerógena de un alimento con contenido elevado en radicales libres.
