Evaluation of 99mTc-ciprofloxacin formulation with reductor SnCl2•2H2O and Sn-tartrat

99mTc-ciprofloxacin is an antibiotic-based radiopharmaceutical for the diagnosis of infections caused by bacteria. In this study, 99mTc-ciprofloxacin was prepared by using two kinds of reducing agents which were SnCl2•2H2O and Sn-tartrate. The aim of this study is to compare each of the reducing agents which produce 99mTc-ciprofloxacin with high radiochemical purity and good accumulation in the infection site. The results of this study showed that the optimum conditions with radiochemical yield 94.50 + 1.60 were obtained at 25 µg SnCl2•2H2O while Sn-tartrate at 400 µg were obtained radiochemical yield 93.28 + 2.26. Radiochemical purity analysis of 99mTc-ciprofloxacin using High-Performance Liquid Chromatography showed the same chromatogram profile for each reducing agent with a peak of 99mTc-ciprofloxacin found at a retention time of 7.17 minutes. Biodistribution study of each reducing agent on thighs of mice infected with Staphylococcus aureus showed that formulation of 99mTc-ciprofloxacin with SnCl2• 2H2O showed a relatively constant value of target / non target ratio in 1 hour and 3 hours post-injection; 2.71 and 2.25 respectively. While the target / non target ratio value of 99mTc-ciprofloxacin with Sn-tartrate reducing agents at 1 hour and 3 hours post-injection was 1.78 and 2.20. From the results of the study it can be conclude that the formulation of 99mTc-ciprofloxacin with SnCl2•2H2O and Sn-tartrate as reductor has different characteristics. (author)
