The effects of X-rays on the proliferation dynamics of cells in the imaginal wing disc of Drosophila melanogaster

Description

The size distribution of clones marked by mitotic recombination induced by several different doses of X-rays applied to 72 h old Drosophila larvae is studied. The results indicate that irradiation significantly reduces the number of cells which undergo normal proliferation in the imaginal wing disc. It is estimated that 1000r reduces be 40-60% the number of cells capable of making a normal contribution to the development of the adult wing. Part of this reduction is due to severe curtailment in the proliferative ability of cells which nevertheless remain capable of adult differentiation: this effect is possibly due to radiation-induced aneuploidy. Cytological evidence suggests that immediate cell death also occurs as a result of radiation doses as low as 100r. The surviving cells are stimulated to undergo additional proliferation in response to the X-ray damage so that the result is the differentiation of a normal wing. (orig./MG)

Die Groessenverteilung von Klonen, die durch mitotische Rekombination markiert wurden, die durch verschiedene Dosen von Roentgenstrahlen bei 72 stuendiger Bestrahlung von 72 h alten Drosophila-Larven herbeigefuehrt wurde, werden untersucht. Die Ergebnisse deuten darauf hin, dass Bestrahlung die Zahl der Zellen, die eine normale Proliferation in der imaginalen Fluegelscheibe durchmachen, erheblich vermindert. Es wird geschaetzt, dass 1000r die Zahl der Zellen, die eines normalen Beitrags zur Entwicklung des ausgewachsenen Fluegels faehig sind, um 40-60% vermindern. Ein Teil dieser Verminderung wird verursacht durch die starke Herabsetzung der Proliferationsfaehigkeit der Zellen, die jedoch trotzdem ihre Faehigkeit der Proliferation im ausgewachsenen Zustand nicht verlieren; dieser Effekt wird moeglicherweise durch strahlungsbedingte Aneuploidie bewirkt. Zytologisch wurde nachgewiesen, dass sofortiger Zelltod auch ein Ergebnis der niedrigen Strahlungsdosen wie 100r sein kann. Die ueberlebenden Zellen werden stimuliert, eine weitere Proliferation durchzumachen, als Reaktion auf die durch Roentgenstrahlung entstehenden Schaeden, so dass das Ergebnis in der Differenzierung eines normalen Fluegels besteht.
